The effects of local anesthetics, procaine and mepivacaine, were examined using isolated rat liver mitochondria as a model membrane system. The effects of procaine and mepivacaine were studied on mitochondrial respiration, calcium stimulated respiration and gramicidin-induced K+ efflux and swelling. High concentrations (about 10^-3 and 10^-4 M) of procaine and mepivacaine were found to stimulate the respiratory rates and decrease the RCRs of mitochondria. High concentrations of procaine and mepivacaine also inhibited the rate of Ca++-stimulated respiration, induced by the presence of both limited and massive amounts of Ca++, under both normel stoichiometric and "superstoichiometric” conditions. There are two types or sheep distinguished by the intracellular potassium level or their red blood cells, i.e. low potassium red cells and high potassium red cells. Studies by other investigators have shown that both the rate or active sodium and potassium transport and sodium- plus potassium-stimulated adenosine triphosphate hydrolysis (Na+,K+-ATPase) are approximately four times greater in high potassium than in low potassium red cells. In 9 patients with chronic bronchitis and obstructive ventilator defects: 1) Perfusion was decreased in poorly ventilated regions, though to a lesser extent than the reduction in ventilation. 2) Four patients (group I) had no significant change in distribution of perfusion while breathing 40% oxygen and possible explanations are discussed. These patients all had severe chronic obstructive ventilatory defects with carbon dioxide retention and 3 had a history of cor pulmonale. 3) Five patients (group II) had a significant tendency to improve perfusion in poorly ventilated regions while breathing 40% oxygen. The feasibility of recording from several neurons in an animal's nervous system was established. A review of previous attempts at multineuronal recording is presented. An improved method for recording simultaneously from several neurons in an in vivo preparation was developed. The approach developed uses one microelectrode to record extracellularly from several neurons in close proximity. It can be assumed that the action potentials, originating from each neuron will exhibit different shapes. Cell-mediated immunity was induced by the injection of a low dose of sheep erythrocytes in Complete Freund's Adjuvant. This state of cell-mediated immunity was found to correlate with a reduce capacity of the spleen cells taken from such animals three days after priming to produce anti-sheep erythrocyte antibody in vitro. Evidence presented suggested that the depression is due to a competition between the cell mediated and humoral responses for the same thymus-derived lymphocyte. Thymus-derived lymphocytes perform a helper function in the synthesis of anti-sheep erythrocyte antibodies in mice. Nuclear magnetic resonance has been used to study the association of atropine and eserine with acetylchol inesterase extracted from squid head ganglia and synaptosome membranes from rat brain cortex. Line width changes of the N-methyl and phenyl groups of atropine and the N-methyl and C-methyl groups of eserine resulting from association with the enzyme and the synaptosome membranes have been used to study the interactions. The results show that whereas eserine binds at the active site of the enzyme, atropine binds at a site distinct from the active center.
