Normal rabbit bone marrow cells, but not cells of the other normal lymphoid organs could transfer immunocompetence to lethally irradiated or tolerant allogeneic recipients. However, bone marrow cells of rabbits immunized 24 hours previously could not transfer immunocompetence with respect to the immunizing antigen. The bone marrow immunocompetent cell was identified as the antigen reactive cell (ARC) and not the antibody forming cell (AFC) by the use of allotypic markers. The ARC which were morphologically small lymphocytes, could be isolated by passage of the normal bone marrow cells through an antigen-sensitized glass bead column. An electron microscopic study of colloid droplets and dense bodies in the thyroid follicular cells of TSH-injected and non-TSH-injected rats indicated the following: 1) Colloid droplets and dense granules were situated on the average slightly nearer to the apex of the cell than to the base. On the average, one micron separated a colloid droplet from the nearest dense body. TSH did not significantly affect these values. 2) Colloid droplets varied in density between gray and white. The purpose of this thesis is to deal with Mu'nis' political and military career at the 'Abbasi court between 296-321/908-933. Three focal areas were chosen for this study. (1) The struggle for power between the military class and the kuttab as exemplified by Mu'nis' struggle with the wazir Ibn al-Furat. (2) The struggle for power within the military class itself as it appears through Munis' struggle with Harun b. Gharib and Yaqut. Calf-thymus and mouse Erlich ascites carcinoma DNA-RNA hybrids were prepared by transcribing denatured DNA in vitro with RNA polymerase. The hybrids were purified by chromatography on methylated albumin kieselguhr after digesting the unhybridized nucleic acid with Neurospora crassa endonuclease. The sedimentation constants of both calf-thymus (11% and 19% pure) hybrids and mouse-tumor (28% pure) hybrid in sucrose gradients were approximately 4S. The hybrids melted at 73 – 76 °c (in 0.1M Tris-HCl, pH 7.5, 0.01M MgCl2 , 0.1M NaCl). By inducing a steady state of 133Xe exchange across the lung and measuring the 133Xe concentration in individual lung regions, we were able to measure steady state regional ventilation/perfusion ratios (VA/Q). This technique was combined with independent assessments of regional ventilation and blood flow, and a variety of subjects studied. Erect normal subjects showed an apex-to-base gradient in VA/Q which, though sizable, did not explain all VA/Q variation reported in such lungs. Seated post-pneumonectomy patients wew examined; the increased flow through the remaining lung decreased, apex-to-base differences in perfusion and VA/Q, but ventilation distribution was unchanged. The incremental-pragmatic world-view offers an approach to political development in the Third World. The analytical orientation of the approach is determined by an underlying value-system. An empirical investigation of India historically and sociologically contradicts the theoretical analysis of this approach. The latter is apparently unable to produce a meaningful understanding of Indian socio-political forces because underlying values distort empirical evidence. The contemporary Indian liberal-democratic political system represents the practical application of the theoretical analysis. For related and similar reasons the practical dimension of the incremental pragmatic approach is evidently unsuccessful. The Merrifield method of Solid Phase peptide synthesis has been applied to a study of the evolution of the neurohypophysial hormones. This work was carried out in an attempt to establish the identity of a principle designated EOP 1 (elasmobranch oxytocin-like principle 1) which had been isolated from the pituitary gland of the spiny dogfish (Squalus acanthias), and to test hypotheses on possible evolutionary intermediates. Three new analogues, 8-glutamine-oxytocin, 8-phenylalanine-oxytocin and 4-proline 8-glutamine-oxytocin were synthesized while a fourth, 8-valineoxytocin, previously synthesized by classical methods was synthesized for the first time by solid phase. Methods of isolation of IgD from D-myeloma and normal sera have been explored as has a sensitive method to quantitate low levels of IgD. DEAE-cellulose chromatography of a 30-45% saturated ammonium sulfate fraction of D-myeloma serum, yielded an IgD rich fraction in which IgG contamination did not exceed 20-25%. This was fractionated on G-200 Sephadex and IgD was eluted free of IgG in the ascending half of the first protein peak. Specific antisera to IgD were insolubilized by copolymerization and by coupling to an insoluble carrier and were used to specifically adsorb IgD from normal sera. Serial arterial blood gas studies and pulmonary diffusing capacity measurements were made in 43 patients admitted with fractures to detect subclinical fat embolism. The values were correlated with chest radiographs, coagulation factors and serum lipid fractions. Eight patients developed fat embolism, with all but one of them having multiple fractures. Significant arterial hypoxemia developed in all of the patients with fat embolism and was detected early following injury. The other tests were of limited value. A faster, more efficient method has been developed for preparation of a copper-protein similar to Mann and Keilin's hepatocuprein. Beef liver is homogenized in glass-distilled water. The water-soluble fraction, prepared by centrifugation at 105,000 g, is heated at 50° for 10 minutes and recentrifuged. Residual soluble protein is precipitated with acetone. On redissolving in 0.005M K-phosphate buffer, pH 7.0, the material is passed through Sephadex G-75 and fractions with a high copper:protein ratio are pooled. These fractions are colorless.
