1971 — A transmission and scanning electron microscopic study of broncho-pulmonary epithelial structures.Résumé
The epithelial cells of the distal airway were studied under four different conditions in mice, rabbits and humans. Evidences observed strongly support the theory that osmiophilic bodies are phospholipid components of pulmonary surfactant and type II cells secrete surfactant. Clara cells are merocrine type secretory cells but are immature at the time of birth and not likely to secrete surfactant. Adrenalin is not tropic to either Clara cells or type II alveolar cells. The presence of cytomegalovirus-like particles within Pneumocystis organisms prompted a hypothesis of symbiosis between these two agents. Regeneration, function, and maintenance of integrity of the liver were studied in five groups of hemihepatectomized dogs with varying hemodynamic inflow to the residual half liver, and one group with intact livers but differentially ligated inflow to each half. Atrophy is distinguished from lack of regeneration, and is shown to be solely dependent upon adequacy of total volume of liver blood flow, and not any trophic substance or specific type of flow. This concept is extended to the problems of revascularization and atrophy of auxiliary liver transplants. The purpose of this experiment was to study cytological changes in the thymus of neonatal mice during hydrocortisone induced involution and regeneration with the light and electron microscopes. Neonatal mice of strain ICR/Ha were used in this study. They were injected subcutaneously with 0.1 mg hydrocortisone within 24 hours after birth. The weights, mitotic indices, and histology of the treated thymuses were compared to the control groups at 6 hours, 24 hours, and 3, 15, and 30 days post-treatment. An in vitro system containing a rabb1t serum medium adapted subline of L5178Y cells and rabbit antiserum was used to study the effects of antibodies, in the absence of complement, on cells in culture. Heat-inactivated antiserum inhibits cell growth and colony formation in soft agar, and kills the cells. These actions occur slowly and metabolic events, in sequence; that is, RNA synthesis is inhibited after 4 hr, but the effects on the rates of protein and DNA syntheses become demonstrable only after 6 to 8, and 8 hr, respectively. The metabolism of free fatty acids in hemorrhagic and endotoxin shock was studied in a group of 93 mongrol dogs. The changes produced by norepinephrine and isoproterenol infusions in the shock state were evaluated. The concentration of FFA in the plasma of dogs anesthetized after a 12-16 hour fast was 450 µEq/l. (range 150-1040 µEq/l). Norepinephrine and isoproterenol increase the plasma FFA concentration by mobilizing FFA from adipose tissue. A moderate hemorrhage increases the plasma FFA concentration, but a several hemorrhage produces no such increase. (1) Dietary manipulation of thyroid function was shown to be associated with changes in the activity of adenyl cyclase and cyclic AMP phosphodiesterase that signify a rapid turn-over of cyclic AMP in the chronically stimulated gland and the reverse in the suppressed gland. Measurement of a fairly constant concentration of cyclic AMP in these circumstances further emphasized the physiological importance of turn-over rate rather than concentration of the nucleotide. 1971 — Distributed models of transcapillary exchange and their use in the study of the heart.Résumé
In Chapter one theoretical indicator dilution curves are calculated using different mathematical models. In Chapter two experimental indicator dilution curves as obtained from working dog hearts were found compatible with a distributed diffusion limited model. The model parameters were obtained by performing least square error curve fits of the model to the experimental data. In Chapter three the above model was extended to include one way cellular uptake. The toxic reaction which can occur when patients taking monoamine oxidase (MAO) inhibitors are given meperidine has never been satisfactorily explained. It is well known that MAO inhibitors elevate brain amines, and compounds such as morphine and meperidine have also been shown to affect these agents. It therefore appeared reasonable that the interaction between phenelzine and meperidine might be linked to their combined effect on one or more of these amines. Biotin deficiency has been shown to cause morphological aberrations in A. globiformis 425. Single compounds, such as oleic acid, linoleic acid, linolenic acid, Tween 80, aspartame, and oxalacetate, showed varying degrees of biotin-replacing activity, but none was able to restore the cell to normal growth. Comparative analysis of fatty acid composition indicated that biotin deficiency causes a shift in synthesis from anteiso to normal straight chain fatty acids. RNA, DNA, and protein accumulation were followed throughout the growth cycle.
