1972 — Control of polyamine synthesis in normal and neoplastic rat liverRésumé
A study has been made of the regulation in normal and neoplastic rat liver of the induction of ornithine decarboxylase, a key enzyme in polyamine synthesis in eukaryotes. The radioassay for this enzyme was significantly improved by the addition of EDTA and the substitution of Tris buffer for phosphate buffer. In contrast to DNA synthesis the profile of the ornithine decarboxylase induction in response to partial hepatectomy does not vary dramatically with the age of the rat. The induction of ornithine decarboxylase is inhibited by 8-azaguanine and 5-azacytidine, inhibitors of protein synthesis. The induction is however reinitiated after a delay … Lire la suite

1972 — The site of binding of iodine to thyroglobulin in rat thyroid follicles as revealed by quantitative electron microscope radioautographyRésumé
After injection of 125I into animals, discovery in radioautographs of some silver grains over cells of thyroid follicles continues to provoke the controversy that binding of iodine to protein might occur in cells rather than in the colloid of follicle lumens (Croft and Pitt-Rivers, 1970; Takano and Honjin , 1968). To analyze the significance of grains over cells, 10 µCi per gram body weight of Na 125I was given intravenously into adult male rats which were sacrificed in 30-45 seconds by perfusing with glutaraldehyde; over 700 photographs of EM radioautographs were examined. Quantitative studies (by drawing appropriate boundary circles about … Lire la suite

1972 — Mammalian tumour tRNA methylasesRésumé
A tRNA N2 guanine dimethylase was partially purified from mouse Ehrlich ascites tumour cells, and some of its properties studied. The enzyme was purified from 105,000g supernatant by precipitation at pH 5.1 followed by chromatography on hydroxyapatite at pH 6.8. Activity was assayed by measuring the rate of methylation of either E. coli B or yeast tRNA with (14C or 3H)-Me-S-adenosyl-, methionine at pH 8.2. Although substantial amount of other methylated guanine derivatives and methylated pyrimidines were detected in tRNA incubated with the "supernatant enzymes" fraction or with the "pH 5 enzymes" fraction, 86% of the methylation of tRNA with … Lire la suite

1972 — Neural correlates of behavior in an auditory discrimination taskRésumé
Multiple-unit activity (MUA) and average evoked potentials (AEP) were simultaneously recorded from rats performing in a "go, no-go" auditory discrimination task. Average MUA was generally less variable and better related to the animal's response than was the AEP. The lateral septal area was the only structure where the poststimulus pattern of MUA was consistently related to whether or not the animal bar pressed. Medial septal prestimulus MUA levels were highest when the animal did not subsequently bar press, while poststimulus MUA levels were always sharply reduced from the prestimulus range, regardless of stimulus or response conditions. These results were frequently … Lire la suite

1972 — Food in the Canadian North and Changing Dietary PatternsRésumé
"The purpose of this monograph is to focus attention on the changing food habits of people living in the Canadian north. In order to understand the enormity of the problems that have traditionally faced people living in this region, the introàuction will give a brief assessment of the environmental factors of the Arctic and subarctic. Observations have been made concerning the traditional life style when hunting was the sole means of subsistence. The process of acculturation which began decades ago through contact with the white man has accelerated dramatically since the Second World War. The housekeeping and health problems of … Lire la suite

1972 — Isolation of biologically active substances from oak galls exerting antihistaminic and anti-inflammatory activity.Résumé
The work presented in this thesis describes a method for the isolation of an antihistamine-like substance (KC-18) from the Hungarian oak gall extracts. Data obtained concerning the structure of KC-18 strongly suggested that it is an ester of piperonylic acid. In animal experiments, KC-18 was found to antagonize the actions of histamine in a dose-related manner when tested by the aerosol and capillary permeability methods. KC-18 also exerted anti-anaphylactic activity in actively sensitized guinea pigs. In addition, KC-18 inhibited histamine-induced hypotension and gastric acid secretion, but it could not antagonize the histamine-induced contraction of the isolated guinea pig ileum. The … Lire la suite

1972 — The structure and metabolism of galactosylgalactosyiglucosylceramide.Résumé
Galactosylgalactosylglucosylceramide (CTH) was purified from normal human kidneys, and the kidney of a patient with Fabry's disease (characterized by a defect in the catabolism of CTH). NMR spectra of the intact molecules indicated the presence of two beta-D-glycopyranosyl residues and one alpha-D-glycopyranosyl residue. CTH and lactosylceramide labelled in the terminal galactose residues were used to show that the terminal galactosidic linkage of CTH, but not lactosylceramide, was susceptible to hydrolysis by an alpha-galactosidase from coffee beans. Labelled CTH and p-nitrophenyl- alpha-D-galactopyranoside were used to demonstrate concomitant deficiencies of CTH galactohydrolase and nonspecific alpha-galactosidase in the kidney of a patient with … Lire la suite
