    Fishing is a potent ecological force that can impact the density and distribution of fish stocks as well as fitness-related traits. In this thesis I examined eco-phenotypic change in the introduced Nile perch of Lake Nabugabo (East Africa) in response to decades of intense fishing pressure. I quantified temporal (12 years) and spatial variation (wetland vs. exposed habitats) in the a) size, b) catch per unit effort, c) diet, and d) gill morphology of Nile perch, by comparing field data collected in 2007 to earlier studies (1995, 2000 for distribution and diet and 1996 for gill morphology).     2009 — Mantle source and petrogenesis of kimberlites from the Foxtrot Kimberlite Field of northern Québec, Canada

    Kimberlite whole rock geochemistry has been under-appreciated in deciphering Group I kimberlite petrogenesis. Although there is considerable debate on the definition of Group I kimberlite, there is agreement on certain characteristics. These include that Group I kimberlites are ultramafic rocks (MgO >15 wt%), have near primitive mantle nickel (~ 1400 ppm) and chromium (~1600 ppm) contents, have close to bulk silicate earth radiogenic isotopic signatures, are volatile rich (predominantly CO2) and have stable isotopic signatures typical of mantle sources.     The finite element method (FEM) is used to solve three-dimensional electromagnetic scattering and radiation problems. Finite element (FE) solutions of this kind contain two main types of error: discretization error and boundary error. Discretization error depends on the number of free parameters used to model the problem, and on how effectively these parameters are distributed throughout the problem space. To reduce the discretization error, the polynomial order of the finite elements is increased, either uniformly over the problem domain or selectively in those areas with the poorest solution quality.     Bio-composite materials suitable for replacing wood for use in musical instruments were developed. The mechanical properties of Sitka spruce, the most widely used wood species for this application, were taken as a benchmark when developing the new materials. The materials were characterized by static and dynamic methods to determine the dynamic Young's modulus, shear modulus, internal friction and static mechanical properties.     2009 — Methylenetetrahydrofolate reductase and dietary folate: developmental impact and gene regulation

    Maternal genetic and nutritional influences have a vast impact on developmental outcome. In particular, disturbances in folate metabolism have been well-studied since the finding that dietary folate dramatically reduces the incidence of neural tube defects (NTD). A mild deficiency in the folate-metabolizing enzyme, methylenetetrahydrofolate reductase (MTHFR), and altered dietary folate are clearly associated with NTD, however, their effects on other congenital defects and pregnancy complications are not conclusive. In this thesis, the impact of MTHFR deficiency and altered dietary folate on embryonic and placental development were investigated in our mouse model of MTHFR deficiency.     Single-channel speech enhancement algorithms are used to remove background noise in speech. They are present in many common devices such as cell phones and hearing aids. In the Bayesian short-time spectral amplitude (STSA) approach for speech enhancement, an estimate of the clean speech STSA is derived by minimizing the statistical expectation of a chosen cost function. Examples of such estimators are the minimum mean square error (MMSE) STSA, the β-order MMSE STSA (β-SA), which includes a power law parameter, and the weighted Euclidian (WE), which includes a weighting parameter. Gene expression profiling has divided this heterogeneity into luminal, ERBB2+ and basal molecular subtypes. Basal breast cancers are difficult to treat as they lack expression of candidates suitable for targeted therapies and are associated with poor outcome.     This thesis investigated the factors governing pond-breeding amphibian life stage occurrences at multiple sites within a 467-hectare forest at Mont-Saint-Grégoire, Québec, during 2006 and 2007. Several environmental, landscape characteristics and predator occurrences were tested using different statistical analyses to predict occurrence of these life stages. Life stages responded to different conditions and were not predicted by the same factors. Recruitment can be successful only if conditions are suitable for all life stages. Recruitment was poorly predicted by the detection of breeding adults, while it was better predicted by that of eggs and tadpoles.     DNA replication initiates at replication origins and involves the ordered assembly of a pre-Replicative Complex (pre-RC), consisting of the ORC1-6, Cdc6, Cdt1 and MCM2-7 proteins. The Ku heterodimer (Ku70/Ku80) was previously implicated in the pre-RC assembly but its mechanism of action is yet unknown. Here, Ku is shown to form a complex with DNA topoisomerase IIβ (topo IIβ) both in vitro and in vivo and target it onto replication origins. The enzymatic activity of topo IIβ creates transient, origin-specific dsDNA breaks thus affecting the DNA topology during pre-RC assembly.     The eIF2α kinases PERK and PKR are well characterized regulators of protein synthesis. When they are activated in response to various cellular stresses, they inhibit translation initiation by phosphorylating eIF2α on a key residue. Recently, PKR has been shown to regulate the expression of specific proteins independent of its ability to inhibit protein synthesis, and we wished to identify additional targets of this particular function.     Spinal cord injury affects thousands of people every year, however, treatment options are currently limited and none are universally accepted. Inflammation has been shown to play a critical role in propagating secondary damage which exacerbates the initial trauma, therefore modulating the inflammatory response after injury has therapeutic potential. As prostaglandins are powerful inflammatory mediators in the periphery, I have examined the role of prostaglandin D2 (PGD2) after spinal cord injury. My thesis provides evidence that PGD2 produced from hematopoietic prostaglandin D synthase plays a detrimental role in locomotor recovery.
