    Childhood blindness due to hereditary retinal disease represents a huge clinical problem. Morbidity, blindness but also a major lack of understanding the molecular genetic events that lead to blindness in very young children hamper clinical management of this important and common problem. One way to get entry into the disease mechanism of childhood retinal blindness is to study Mendelian disease entities that are caused by relatively straightforward links between mutations in genes that lead to aberrant retinal protein function and the resulting disease phenotype. The present research project contributes to the development of an injectable biomaterial for potential use in treating vocal fold (VF) lamina propria (LP) scarring disorder. A new biomimetic hydrogel system, based on hyaluronic acid (HA) and gelatin (Ge), with hierarchical structural organization was designed, characterized, and evaluated in vitro and in vivo. This hierarchical composite hydrogel was constituted from hybrid HA–Ge microgels, fabricated through water-in-oil mini-emulsification polymerization, embedded in a secondary HA network.     The stringent response (SR) is a conserved bacterial stress response mechanism that allows bacteria to survive during stress and starvation. This response is mediated by (p)ppGpp, a hyperphosphorylated guanosine signal that regulates expression of genes to inhibit cellular proliferation and induce genes important for survival and adaptation. This study investigates the role of the SR in regulating oxidative stress pathways in Pseudomonas aeruginosa (PA), a human opportunistic pathogen that can cause infections in immunocompromised patients, and patients suffering from cystic fibrosis.     This thesis examines the experiences of the first wave of Pakistani immigrants to arrive in Canada after the immigration reforms of the 1960s and 1970s. It focuses largely on the Pakistani community in Toronto. Its primary thesis is that while the immigration reforms and accompanying national policy of multiculturalism of this era were intended to foster democratic, humanitarian, and egalitarian principles, Pakistanis, like other immigrants of colour, actually saw their efforts to join the social and economic mainstream thwarted by discrimination both at the hands of government officials and within Canadian society.     Histone deacetylase inhibitors (HDACi) are anti-cancer agents, which have shown promising activity in hematological malignancies. However, only a small percentage of patients initially respond to treatment and all will eventually develop resistance. HDACi are known to inhibit deacetylation of histones as well as various non-histone proteins.     The Bowone and Binebase Au (Ag-Cu) deposits are related prospects, located 1 km apart on the volcanic island of Sangihe, Northeastern Indonesia. The deposits occur at the southern and oldest end of the island, and are hosted in Quaternary volcanic and volcaniclastic formations. Each deposit has a supergene and hypogene mineralized zone, although only the supergene zone of the Binebase deposit is of significant thickness. As part of a new lifecycle approach to drug regulation, Health Canada recently announced plans to develop a new health product vigilance framework that will allocate drug safety resources using prioritization schemes focused on higher risk.     This thesis examined the Simple View of Reading (SVR; Hoover & Gough, 1990) in predicting writing performance. Specifically, the present study measured whether unique variance is explained by both decoding and language-related variables in predicting sixth-grade writing performance. The contribution of working memory in predicting writing was also examined, guided by the multi-component model of working memory (Baddeley, 2000).     This thesis describes our advances in the preparation of lipophilic fluorogenic probes for the specific imaging of reactive oxygen species (ROS) in the lipid membrane of live cells. The work revolves around a simple design involving the preparation of a two segment receptor-reporter type free radical scavenger-fluorophore probe (an off-on fluorogenic antioxidant indicator). The receptor segment in the probe mimics the structure and activity of the naturally occurring antioxidant α-tocopherol.     The original goal was to use a downcomer to create swarms of solvent-coated bubbles to scale-up the air-assisted solvent extraction (AASX) process. The downcomer provides a close-packed bubble bed considered attractive for solvent coating and fast extraction kinetics. During initial testing, the bubble bed was observed to collapse upon introduction of solvent, evident from decreasing gas holdup, decreasing aspirated air rate, and formation of air slugs. Neither changing solvent introduction technique, solvent composition nor addition of frother or salt alleviated the situation.     Background Temporal lobe epilepsy (TLE) is the most common drug-resistant epilepsy in adults. Hippocampal sclerosis, the hallmark epileptogenic lesion of TLE, is characterized by neuronal loss and gliosis. The hippocampus is composed of cytoarchitechtonically distinct subfields which are differentially affected in TLE. Indeed, neuronal loss is observed primarily in the CA1 and to a lesser degree in the subiculum, CA3, CA4 and dentate gyrus. Purpose. Our goal was to develop an anatomy-driven manual hippocampal subfield segmentation protocol on sub-millimetric MRI.     Arsenic trioxide (ATO) is an effective treatment for acute promyelocytic leukemia (APL). While it is undergoing clinical trials for numerous malignancies including multiple myeloma, myelodysplastic syndrome, lymphoma and solid tumors, it has demonstrated only limited efficacy as a single agent. However, it may hold promise as part of a combination therapy.     Using procedural narrative generation in video games provides a flexible way to extend gameplay and provide more depth to the game world at low cost to the developers. Current examples of narrative generation in commercial games, however, tend to be simplistic, resulting in repetitive and uninteresting stories.     Cell death and inflammation are two central, tightly coupled, innate immune processes. The inflammasome triggers both in response to pathogens and tissue damage. This cytosolic signalling complex contributes to host-defence, tissue repair and inflammatory disease, but the factors governing inflammasome activity are poorly characterized. This thesis examines the function of inflammasome modulators in infection and immunity. Inflammasome activity is antagonized by caspase-12. In humans, caspase-12 expression is limited to populations in geographical locations that overlap with regions endemic for malaria.
