2022 — Recombinant protein-based scaffolds: extracellular secretion, purification, and mineralization

2022 — Development of a method for MRI based prediction of microstructural features of the brain
    Résumé

This thesis presents a deep learning framework that aims to learn and predict microanatomical features from magnetic resonance imaging (MRI) of the rat brain. We obtained different MRI contrasts in an effort to understand the underlying mechanisms of MRI signals and their relationship to microstructural features. Scientists have studied numerous neurological disorders through analysing the brain’s anatomy and functionality. For decades, the gold standard for accurately assessing and quantifying microstructural biomarkers of several pathologies and disorders in the brain remained to be histology. However, since acquiring histological images is only done post-mortem, this results in very limited capacity of diagnostics …  Lire la suite

2022 — Inference latency modelling of BERT on ARM cortex-A multi-core processors

…  Lire la suite

2022 — Assessment tool for widespread screening of anterior cruciate ligament injury risk

2022 — The synthesis of 2'-modified RNAs for probing enzymes implicated in splicing and gene editing
    Résumé

The exponential developments made in the field of mRNA vaccines, CRISPR technology and antisense therapies have solidified nucleic acid therapeutics’ presence at the forefront of modern medicine. With an ability to engage targets that are otherwise undruggable by conventional therapeutics, nucleic acid-based drugs have opened new avenues for treating intractable diseases. Synthetic oligonucleotides are now recognized as the third major drug discovery platform, in addition to small molecules and antibodies. Additionally, they provide valuable probes to study key structure-function relations and offer mechanistic insights at the molecular level of important enzymatic processes. In this thesis, we highlight our efforts in …  Lire la suite

2022 — COPD and heart failure comorbidity from patients to mechanisms
    Résumé

Introduction:Chronic Obstructive Pulmonary Disease (COPD) and Chronic Heart failure (CHF) are prevalent chronic conditions. More patients with COPD and CHF have preserved ejection fraction(HFpEF) compared to CHF with reduced ejection fraction (HFrEF). Cigarette smoking, the most important risk factor of COPD, induces systemic inflammation and could lead to the development of cardiovascular disease. We hypothesized that patients with COPD and CHF will differ in lung structure, severity of airflow obstruction and blood biomarkers and that cigarette smoke affects the activation of lung cells. The main objectives of the thesis were 1) to determine the prevalence of co-morbid CHF in COPD …  Lire la suite

2022 — An investigation of proton pump inhibitors: Addressing their utilization and association with gastrointestinal cancers

Tous …  Lire la suite

2022 — The rule of law in mainstream development: an inflated concept?
    Résumé

The Rule of Law is taken to express a straightforward idea: societies should be governed by laws rather than the whims of humans. More precisely, the Rule of Law expresses the idea that the State or sovereign exercises its power within the scope of limiting public norms, namely laws. However, its usage has been taken well beyond its traditional role of curtailing the power of the State or sovereign; it is inserted as a response to virtually every demand of the mainstream development agenda. Indeed, the Rule of Law has been overexpanded by an ever-growing list of development demands and …  Lire la suite

Background: Cerebral palsy (CP) is a lifelong disorder of posture and movement and is the most common cause of physical disability in children. Since there is no cure available for CP, understanding causal mechanisms is essential to develop effective primary prevention strategies. Whereas plenty of research has examined temporal trends in overall CP rates, changes in CP rates by key sociodemographic characteristics over time have been scarcely described. Furthermore, limited evidence exists in the literature on the role of preconception and antenatal modifiable factors that lie early in the causal pathways that lead to CP. Objectives: This doctoral research aimed …  Lire la suite

2022 — Guidance and model predictive control of a tandem-rotor helicopter
    Résumé

The tandem-rotor helicopter is a platform of increased interest for autonomous development. This thesis considers the guidance and model predictive control (MPC) of a tandem- rotor helicopter. First, the dynamics of the tandem-rotor system are developed and a simplified dynamic model is presented. An error definition is formulated using the matrix Lie group SE2(3), which is used to linearize the system. Next, a computationally simple, two-stage guidance strategy is presented. A coarse reference trajectory to a target is calculated using a quartic guidance law, leveraging the differentially flat properties of the system. The trajectory is then refined using a finite-horizon …

2022 — Generation of novel Zasp52 mutants and analyzing the associated phenotypes, and identifying the interaction domain of α-actinin with Zasp52 extended PDZ domain
    Résumé

Striated muscles conduct many essential aspects of life such as moving and heart beating. Sarcomere structure and function as the smallest contractile unit of muscles, is highly conserved throughout evolution. It is composed of actin thin filaments and myosin thick filaments which are confined between two adjacent Z-discs. The Z-disc is a multi-protein complex that serves as an anchor-site for actin filaments. Different Z-disc cross-linker proteins such as α-actinin, filamin and Alp/Enigma proteins are involved in myofibril assembly and maintenance. The Alp/Enigma family is a class of Z-disc-associated proteins, whose mutations lead to myopathies and different muscle disorders. In Drosophila, …  Lire la suite

2022 — Small-angle attraction in the tilt illusion
    Résumé

The tilt illusion defines the phenomenon in which a surround or inducer grating of a particular orientation impacts the perceived orientation of a central test grating. Typically, inducer-test orientation differences of 5-40 deg drive the test grating orientation to appear shifted in a direction away from that of the inducer orientation, i.e., show repulsion. The inducer typically causes the test grating orientation to appear shifted towards that of the inducer orientation in the region 60-90 deg, i.e. shows attraction. Both repulsion and attraction effects have been observed in contrast-modulated and luminance-modulated grating patterns. In this thesis, I demonstrate that a …  Lire la suite

2022 — Signaling events modulated by the exosome-FPR2 axis in early L. major infections
    Résumé

Leishmania is a protozoan parasite that causes leishmaniasis, infecting millions of individuals every year. The parasites utilize different virulence factors to camouflage their presence from macrophages and promote their survival. In recent years, evidence has emerged for the role of exosomes as virulence factors. Co-inoculating mice and macrophages with L. major exosomes exacerbate infections and increase parasite load. Our lab recently identified a potential role of Formyl Peptide Receptor 2 (FPR2) in exosome-mediated exacerbation of L. major infections. Hence, in this thesis, we investigate the interaction between FPR2 and L. major exosomes, and we evaluate downstream signaling pathways activated by …  Lire la suite

Epithelial cells interact to form architecturally organized structures in tissues, with strict control of motility, proliferation and metabolic signaling, each of which is altered during cancer progression. Loss of epithelial organization and tissue architecture is associated with the acquisition of a malignant phenotype during breast cancer initiation and throughout its progression. The changes in tissue architecture that accompany the development of breast cancer is an excellent model, that develops through architecturally different preinvasive stages that include benign lesions such as ductal carcinoma in situ (DCIS), which are risk factors for the development of invasive disease (IDC). Currently, DCIS accounts for …  Lire la suite

2022 — Contributions of Kinesin V to establishing a minimal metaphase spindle length required for chromosome biorientation
    Résumé

In all eukaryotic cells, duplicated chromosomes are segregated into daughter cells by the mitotic spindle. As cells enter mitosis, the spindle forms, and the process of attaching sister chromatids to spindle microtubules continues throughout metaphase. Microtubules attach to a complex of proteins called the kinetochore, located at the centromere of each sister chromatid. Chromosome biorientation occurs when the kinetochores of both sister chromatids are attached to microtubules originating at opposite spindle poles. Chromosome biorientation in metaphase is followed by chromosome segregation in anaphase and finally cytokinesis. In animal cells, chromosome biorientation requires many microtubules to bind to each kinetochore. In …  Lire la suite

    Résumé

Brain organoids are three-dimensional multicellular structures capable of faithfully recapitulating the in vivo brain. Brain organoids grown from human induced pluripotent stem cells constitute promising tools for investigating mechanisms that underlie human neurodegenerative diseases and neurodevelopment. A major challenge that hinders applications of brain organoids is their lack of vasculature, which leads to cell death at their core due to insufficient delivery of oxygen and nutrients once they exceed the diffusion limits that enable oxygenation and nutrient supply. Attempts to vascularize brain organoids often include differentiation protocols performed in traditional well plates, which do not provide a straightforward method for …  Lire la suite

Once the AhR is activated, it translocates into the nucleus, dimerizes with the AhR nuclear translator (ARNT), and binds to specific DNA sequences called xenobiotic response elements (XRE) to promote the target gene transcription, including cytochrome P450 (CYP1A1). The AhR not only mediates the toxic effects of dioxin, but also has physiological functions such as suppressing inflammation, oxidative stress, and apoptosis. We have previously shown that AhR-dependent regulation of endogenous miR-196a expression …  Lire la suite
