1977 — Purification by bioaffinity chromatography of two arginine-specific periplasmic binding proteins.Abstract
Two arginine-specific periplasmic binding proteins have been purified from E. coli ML-30 by affinity chromatography on an arginine-BE-Sepharose 48 gel. By this means it was possible to separate the arginine-binding activity of crude osmotic shock fluid into arginine specific (adsorbed) and arginine non-specific (non-adsorbed) binding frictions. The adsorbed fraction which was displaced with arginine was resolved into two arginine-specific binding proteins by gradient elution with arginine. Both proteins differ on the basis of affinities for arginine, Isoelectric Points, mobilities on anionic polyacryilimide gels, but have identical molecular weights, and similar amino acid composition. Bacterial luciferase, a heteropolymeric protein (alpha,beta) catalyzes the conversion of chemical energy into light. Information about the mechanism of bioluminescence can be drawn from the elucidation of amino acid residues involved in the active site of luciferase. It has been shown that the histidyl residues of luciferase can be specifically modified at pH 6.1 with ethoxyformic anhydride with a concomitant loss of bioluminescent activity. The transport of tryptophan (TRP), precursor of 5-hydroxytryptamine, (5-HT or serotonin) in the central nervous system, was investigated in brain areas known to vary in their number of serotoninergic cell bodies and nerve endings. The regional transport of TRP at the blood-brain barrier was studied with the Oldendorf technique. The passage of TRP across the cell membrane of neurones and glia was studied in slices prepared with a McIlwain tissue chopper, and with isolated nerve endings (synaptosomes) partially purified on density gradients.
