    DNA immobilization was demonstrated using the emulsification/internal gelation technique for calcium alginate bead production. The microspheres formed were recoverable magnetically, resistant to acidic environments, and strong enough to withstand gastrointestinal transit. Purification of DNA from alginate was accomplished sing a Sepharose CL-2B gel column, resulting in decreased alginate interference when quantifying DNA by absorbance. DNA microspheres were studied in vitro for the ability to trap ($ sp{14}$C) methyl iodide and in vivo with ($ sp{14}$C) benzo(a)pyrene.     Alginate beads produced by both external and internal gelation techniques were Studied so as to determine the optimal bead matrix structure within which DNA can be immobilized for in vim application. Alginates were characterized for guluronic/marmuronic acid (G/M) content and average molecular weight. Non-homogeneous calcium, alginate and DNA distributions were found within gels made by the external gelation method because of the external calcium source used.     The N-terminal SH2 domain of protein tyrosine phosphatase 1C (SHN1C) was expressed in E. coli as a fusion to maltose-binding protein (MBP) using two different polylinkers, one comprising of the traditional recognition motif for FXa (IEGR) and the other for thrombin (FNPR), as part of an effort to resolve its three-dimensional solution structure by NMR spectroscopy. The construct encoding the thrombin cleavage site provided a fusion protein that exhibited superior proteolytic cleavage efficiency. This was accomplished using site-directed mutagenesis, kinetic measurements, the identification of processing intermediates, and the analysis of the various X-ray crystal structures reported to date.
