    2020 — The role of mitochondrial proteins CHCHD10 and CHCHD2 in neurodegenerative disease

    Dysfunctional mitochondria have been suggested to contribute to a number ofneurodegenerative diseases including some of the most prevalent such as amyotrophic lateralsclerosis (ALS) and Parkinson’s disease (PD). Our studies focused on two proteins, CHCHD10and CHCHD2, which have been linked to ALS and PD, respectively. CHCHD10 and CHCHD2are paralogues, belonging to the CX9C family of proteins that primarily localize to themitochondrial intermembrane space.     ABSTRACTSalivary hypofunction, also known as xerostomia, is a major side effect of radiotherapy for head and neck cancer patients. Current treatments of irradiation (IR)-injured salivary glands (SGs) remain palliative. Our previous studies showed that mouse bone marrow cell extract (BMCE) could restore the secretory function of IR-injured SGs and that native proteins in BMCE were the main effective ingredients for the treatment. However, BMCE therapy requires multiple injections and protein denaturation is a concern during BMCE storage.     Background: Sodium-glucose co-transporter 2 (SGLT-2) inhibitors are the newest class of antidiabetic drugs and have been shown to have cardiovascular benefits. However, numerical imbalances in breast cancer events were observed with certain SGLT-2 inhibitors (i.e., dapagliflozin) in premarketing trials, all occurring within one year of randomization.     The vitamin folate is involved in one-carbon metabolism and the remethylation of homocysteine to methionine. Disturbances in folate metabolism can result in an increased risk of neural tube defects (NTD), other birth defects or pregnancy complications. The most common cause of genetic folate deficiency is the MTHFR 677C>T single nucleotide polymorphism (SNP), which results in elevated levels of homocysteine and a thermolabile methylenetetrahydrofolate reductase (MTHFR) enzyme with reduced activity. Another methyl donor for homocysteine remethylation is betaine, which is derived from the nutrient choline.     This thesis explores the intersections between global finance capitalism and settler colonialism. It argues that, in the mid-to-late nineteenth century, the Canadian public debt played a key role in the onset of settler colonialism. This study concludes with a focus on Epekwitk, the British named Prince Edward Island, as a way to conceptualize both the local and the global aspects of the role of the public debt in settler colonialism.
