The role of adenosine in modulating synaptic transmission in the cat superior cervical ganglion was investigated in this thesis. The first study showed that perfusion of ganglia with exogenous adenosine increased their acetylcholine (ACh) content. The effect was reduced by blockade of nucleoside transport, as if adenosine's action was mediated at an intracellular site. Isotopic labelling of the extra ACh with radiolabelled choline showed that the additional transmitter was due to increased ACh synthesis, and associated with increased choline transport. Embryonic development relies on programmed apoptosis to shape tissues, particularly in the urogenital system (UGS). Our study unveils the crucial role of non-professional efferocytosis in precisely eliminating apoptotic cells during the remodeling of the common nephric duct (CND), a vital process for UGS maturation. A computational model estimates the epithelial efferocytosis process at approximately 9.3 hours. Further investigations highlight the coordination between non-professional efferocytosis, actomyosin contractility, and apoptosis in driving proper CND shortening. The properties of tubulins from Brugia malayi, B. pahangi, Nippostrongylus brasiliensis and rat brain were compared. Tubulins from all nematodes and rat brain were partially purified by polylysine agarose chromatography, those of brain also by cycles of assembly/disassembly, and all by taxol-induced assembly. The tubulins were compared with respect to concentration ($ mu$g tubulin/mg soluble protein), drugs binding and isoforms. The tubulins of B. malayi and B. pahangi were similar. The nature of shape change and aggregation of human and rabbit platelets was assessed by light transmission studies of stirred platelet suspensions and phase contrast microscopy. Platelet shape change and aggregation were differently affected by experimental conditions: pCO(,2), pH and anticoagulants, according to choice of aggregating agents. Optimal physiological conditions were established for functional studies. A new class of potent antithrombotic agents, BL-3459 and BL-4162A, were identified as selective low Km cyclic 3':5'-adenosine monophosphate (cAMP) phosphodiesterase inhibitors.
