2013 — Multiple roles of DDX17 in human immunodeficiency virus type 1 replicationRésumé
Human immunodeficiency virus type 1 (HIV-1) is a small retrovirus that highly depends on the host cell machinery to replicate by completing its lifecycle and producing new infectious viral particles. The complexity of HIV-1 life cycle regulation only reflects the diversity of the virus-host interactions. Cellular helicases are enzymes involved in every step of nucleic acid metabolism, through rearranging ribonucleoprotein complexes. The understanding and the importance of helicases in HIV-1 replication started to emerge a decade ago. Since then, many studies reported the promoting or inhibiting effect of this protein family on HIV-1. My thesis project was to investigate the … Lire la suite

2013 — Optimizing the representation of space in the parietal cortexRésumé

2013 — Clinal variation and phenology in two conspecifics of Phragmites australis (Cav.) Trin. Ex Steud in Northeastern North America: implications for current management and future climate changeRésumé

2013 — Poly (ADP-ribose) glycohydrolase (PARG) is a new therapeutic target for breast cancerRésumé
Breast cancer is a heterogeneous disease, but is invariably associated with genome wide deregulation of transcription. The silencing of tumor suppressor genes (TSGs) in particular is thought to be an early, initiating event in many cancers. While re-expressing these genes is a clinically relevant goal, the development of therapeutics targeting the reactivation of TSGs has been hampered by a lack of understanding of the mechanisms responsible for TSG silencing. Recently, however, we have described a novel means through which several TSGs become silenced in cancer, which may highlight new therapeutic targets. This involves the epigenetic regulatory factor Ctcf which is … Lire la suite

2013 — SOA modelling and passive wavelength-striped mapping for RODIN architecture in optical interconnection networksRésumé
In order to solve the high demand problem for high data load in communication between data centers, a multi-plane optical interconnection network architecture (RODIN) is suggested for optical interconnection networks [1], in which the scalability and energy-efficiency are emphasized. Since semiconductor optical amplifiers (SOAs) act as energy-proportional routing gate, a dynamic model of SOA is developed to investigate the SOA cascadability and how it will affect the optical signal and communication system performance. The SOA dynamic model is based on the steady-state wideband SOA Connelly model [2], which is chosen due to the precise calculation of material gain from quantum … Lire la suite

2013 — Human adenovirus type 5 E4orf4 interacts wih the nucleopore component nup205 to regulate viral gene expression and replicationRésumé

2013 — Cell cycle uncoupling, elimination, and functional modification of centrioles during C. elegans developmentRésumé

2013 — Schenkerian performance and analysis in dialogue: A reconsideration of Beethoven's Piano Sonata in E minor, Op. 90, iRésumé
Heinrich Schenker's analytical essays in Das Meisterwerk in der Musik and Der Tonwille demonstrate a prescriptive and analytically-biased approach to performance. His critical editions and personally-annotated scores reveal a similar approach, where only the universal structure of a piece—attained through analysis and a consideration of the composer's intentions—should inform performance. Schenker's restrictions on the performer's agency and personal interpretation stand opposed to current scholarship, where a reciprocal and equal relationship between performers and analysts is central. In order to ameliorate this disjuncture, this thesis negotiates between Schenker's polemics and performers' interpretive freedom through the shared concept of ambiguity. Since an … Lire la suite

2013 — Understanding the effects of exposure to an environmentally relevant mixture of brominated flame retardant congeners on the function and development of the male gonadRésumé

2013 — Imagining the city of festivals: festivalization and urban space in MontréalRésumé
This thesis explores the concept of festivalization as it applies to the situation in Montréal, with particular attention to the domain of everyday life. As cities around the world find themselves pressed to respond to economic troubles by developing continuous events, festivals, and other cultural tourism attractions, scholarly examinations have begun to address this phenomenon as "festivalization." However, many studies tend to view contemporary urban festivals as mostly over commercialized, spectacular, and inauthentic, and by extension festivalization as an erosion of a pre-existing everyday life. In response, I argue for the importance of understanding the ways in which festivals participate … Lire la suite

2013 — Understanding human impacts on the phosphorus cycle: implicatons for agronomic and environmental management at multiple scalesRésumé
Modern agriculture has led to fundamental changes in the phosphorus (P) cycle that pose agronomic and environmental challenges at all scales. Phosphorus is a non-renewable resource that is critical to food production because of its role as an essential plant nutrient. At the same time, P runoff from agricultural systems contributes to water quality degradation worldwide. Recent research has begun to illuminate the dimensions of P use in agriculture and its broader sustainability implications, yet there is limited understanding of how these disparate issues are connected across scales. In this thesis, I explore important knowledge gaps related to spatial and … Lire la suite

2013 — Haunted by the gothic: deconstructing the new St. Mary's Anglican church, Walkerville, OntarioRésumé
Ralph Adams Cram (1863–1942) was among the rare Gothicists who practiced both Gothic architecture and literature. He designed several Gothic-Revival churches and campuses across North America, and he wrote a book of Gothic ghost stories in 1895, calling the collection Black Spirits and White. Traditionally, scholars have assumed that the discourses of modern, Anglo-Gothic architecture and literature parted company after the 1830s. Scholars have based that assumption on two interrelated arguments. First, the Victorian Gothic novel evolved beyond the distinctly medieval; whereas, Victorian Gothic architects became rigorously attentive to structural and cultural principles of the Middle Ages. Second, and more … Lire la suite

2013 — Being deaf in a Yucatec Maya community: communication and identity negotiationRésumé
My dissertation sheds light on multilayered experiences of identity in an indigenous, Yucatec Maya community where both deaf and hearing persons use sign language. Owing to the history of Spanish colonialism in this region, and as a result of state approaches which see deafness as pathology, the peoples of Chican acquiesce to assumptions about their identity without necessarily emulating these assertions within community life. At the time I carried out fieldwork, identity assertions in Chican appeared to be reactive, therefore ephemeral, rather than based on some inherent essentialized quality. This was the case for deaf members of the community, and … Lire la suite

2013 — The liability of global navigation satellite system (GNSS) used for air navigation in BrazilRésumé

2013 — The "cheap unseemly, and readily despised" one: a rhetorical understanding of Blandina's gendered performance in «The Martyrs of Lyons and Vienne»Résumé

2013 — A cold cruel teacher: masochistic submission to the films of Michael HanekeRésumé
This thesis offers analyses of two of Michael Haneke's most provocative films, *Funny Games US,* and *The Piano Teacher.* Contrary to the popular critical conclusion that Haneke is a cinematic sadist, this thesis argues that the aesthetics and thematic concerns of his films are best understood through the dynamics of masochism, not sadism. Using a variety of theories of masochism, including the conflicting theories of Sigmund Freud and Gilles Deleuze, this project seeks to build upon the work of Gaylyn Studlar and argue for the subversive power inherent in masochistic submission in the cinema. 2013 — Recovery of PEX1-Gly843Asp associated peroxisome dysfunction by flavonoid compounds in fibroblasts from Zellweger spectrum patientsRésumé
Zellweger spectrum disorder (ZSD) is due to defects in any one of 13 PEX proteins, encoded by PEX genes, which are required for peroxisome biogenesis. ZSD features neurologic, hepatic and renal abnormalities; however, one common mutation, PEX1-p.Gly843Asp (G843D), confers a milder phenotype and represents 30% of all ZSD alleles. Thus, identifying treatments for this allele will benefit many patients. We recently showed that PEX1-G843D behaves as a misfolded protein amendable to improved function by chemical and pharmacologic chaperones. In a small molecule screen, we reported recovery of peroxisomal matrix enzyme import by acacetin diacetate, a flavonoid that may bind to … Lire la suite

Currently, effective treatment strategies for patients with severe chronic obstructive pulmonary disease (COPD) are limited. A widely used treatment method is exercise rehabilitation, which has a multitude of benefits on quality of life, improving exercise tolerance and decreasing hospitalizations. However, patients with more severe cases of COPD are unable to train at exercise intensities high enough to significantly improve disease-related skeletal muscle abnormalities such as losses in muscle mass and strength. Our group has recently shown the feasibility and safety of an eccentric (ECC) cycle training intervention in COPD as an alternative modality that can load the muscle four times … Lire la suite

2013 — On the prediction of power outputs in a microbial fuel cell employing Escherichia coli K12 as the biocatalystRésumé
The lack of access to clean electricity and water in developing nations has given importance to the development of low-cost, widely applicable energy technologies. Microbial fuel cells are being explored as potential sources of clean electricity. A microbial fuel cell (MFC) is a device, in which bacteria produce electrons by oxidizing organic material, which are shuttled from the anode to the cathode, producing a current; the only byproducts of this process are respiratory waste in the form of water and carbon dioxide. Although significant advances have been made in optimizing MFCs for power output, power outputs are not always reproducible, … Lire la suite

2013 — The epithelial cell adhesion molecule (EpCAM) regulates cell motility and cell-cell adhesion by inhibiting PKC signalingRésumé
Tissue cohesion is achieved in part by a large family of plasma membrane-bound cell adhesion molecules (CAMs). During morphogenesis, CAM-mediated interactions provide adhesive forces required for cells to aggregate and form tissues. CAM-mediated adhesions in developing cells are highly dynamic, which provides the fluidity required for cellular movements that drive morphogenesis. Xenopus laevis gastrulation is an established model to study morphogenetic movements. During this phase of development, the mesoderm moves inside the embryo through involution, and migrates along the inner surface of the ectoderm while remaining separated from this tissue. Members of the Fagotto lab have identified the Xenopus orthologue … Lire la suite

2013 — Determining cell-specific gene expression in two soybean mutants using laser capture microdissectionRésumé
Soybean, Glycine max, (L.) Merr., is usually covered in trichomes and has three leaflets per compound leaf. Two mutant soybean cultivars, one without trichomes, cv. Glabrous, and the other with five leaflets per compound leaf, cv. 5-LF, are compared with a wild type cultivar to detect gene expression differences. Trichomes develop and differentiate from the epidermis and the fate of leaves, whether they are compound or simple, is decided in the meristem. Cell-specific gene expression of the epidermis as compared to the meristem is investigated in the three cultivars using Laser Capture Microdissection and high-throughput RNA sequencing. The results indicate … Lire la suite

2013 — Shallow, old and hydrologically inactive fault zones in the Appalachian orogenRésumé
Understanding and quantifying fluid flow patterns and the permeability architecture of fault zones is crucial for petroleum development, the safety of nuclear waste repositories, and water resource management. Fault zones have complex permeability patterns that lead to faults acting as hydraulic barriers, conduits, or combined conduit-barriers. How faults affect groundwater flow at a regional scale (1-10km) is especially uncertain. The objective of this work is to determine whether faults affect regional patterns of groundwater discharge to lakes. We use naturally occurring geochemical tracers to quantify the groundwater discharge to lakes underlain by faults and not underlain by faults, and used … Lire la suite

2013 — The therapeutic potential in eukaryotic mRNA translationRésumé
Inhibitors of translation have proven invaluable in delineating the overall mechanism of protein synthesis. Unlike inhibitors of prokaryotic protein synthesis, the therapeutic development of drugs that directly interfere in the eukaryotic mRNA translation for treatment of human disease has remained largely unexplored. To begin to investigate this possibility and expand the current repertoire of compounds that affect eukaryotic translation, we have undertaken several different experimental screening approaches, two of which are described below and will form the basis of this thesis. In chapter 2, we performed a multiplexed high-throughput chemical screen to identify novel inhibitors of eukaryotic protein synthesis. We … Lire la suite

2013 — Single-molecule fluorescence studies of HIV-1 reverse transcriptase/nucleic acid interactionsRésumé
The human immunodeficiency virus type-1 (HIV-1) is an epidemic affecting approximately 34 million people worldwide. The HIV-1 enzyme reverse transcriptase (RT) plays an essential role in the viral replication cycle by catalyzing the conversion of HIV-1's single-stranded RNA genome into double- stranded DNA. RT is highly dynamic and possesses both DNA polymerase and ribonuclease H activities. While the biologically relevant form of RT is a heterodimeric enzyme consisting of a 66 kDa subunit (p66) and a 51 kDa subunit (p51) (i.e., p66-p51), both p66-p66 and p51-51 homodimers are also catalytically active. The 1812 version of Dominique Vivant Denon's libertine tale Point de lendemain is generally considered to be the second edition of his 1777 original. It is the most frequently published version, cited as the more accomplished one, without particular consideration of the differences between the two versions. However, in 1812 Denon rewrote the tale in an entirely different social, historical and political context, and its significance is thus completely distinct. Furthermore, Denon modified certain aspects of the text, and these changes prove to be significant when analyzed in light of the new historical context.
