Initialement, les composants CRISPR étaient introduits dans les embryons préimplantatoires par micro-injection directe dans le cytoplasme des zygotes ou dans les pronoyaux. Plus tard, la méthode d'électroporation des zygotes a été introduite, qui est simple, à haut débit et conduit à une bonne survie des embryons, mais est limitée à de courtes séquences d'ADN, limitant ainsi le potentiel de CRISPR. The recent cases of Myanmar, Syria and Ukraine suggest that the international community has been failing to implement the third pillar of the ‘Responsibility to Protect’ (R2P) principle in mass atrocity situations. An analysis of these cases demonstrates that the veto power of the permanent members (P5) of the UN Security Council is a significant impediment to its implementation. Developmental and epileptic encephalopathies (DEEs) are a heterogenous group of epilepsies in which altered brain development leads to developmental delay and seizures, with the epileptic activity further negatively impacting neurodevelopment. Identifying the underlying cause of DEEs is essential for progress toward precision therapies. Here we describe a group of individuals with biallelic variants in DENND5A and determine that variant type is correlated with disease severity.
